Objectives: Previous research has shown that probiotics can improve oxidative stress and injury conditions in the kidney tissue. The aim of this research is to determine the antioxidative activity of orange water kefir to improve oxidative stress and the Fraction Area of degeneration and necrosis in the kidney tissue of the hyperlipidemic rats model. Methods: This research used 30 rats divided into 3 groups (K+, K-, B). All groups were fed ad libitum for 8 weeks. The K+ and B groups were given quail egg yolk, 5 ml/200 gr body weight (grBW) for 4 weeks to make a hyperlipidemia condition. Group B was given an orange water kefir with a dose of 5 ml/200grBW. Orange water kefir was made by good manufacturing product (GMP) standards and the procedure for making water kefir. All of this intervention was administered to rats with the sonde method. At the end of this research, all rats were terminated to take the kidney organ to measure the oxidative stress (MDA & SOD) in the kidney tissue, and were dissected transversally, then stained using hematoxylin and eosin (H&E). The Fraction area of degeneration and necrosis was measured by the ImageJ application. Data were collected and analysed with statistical software. Results: After orange water kefir intervention, the mean of MDA level (nmol/gr) was 2.6±0.14 (K-), 15.2±0.13 (K+), and 5.5±0.21 (B). The Mean of SOD activity (%) was 79.23±1.25 (K-), 36.13±2.14 (K+), 59.35±1.13 (B). The mean of fraction area (%) was 0.18±0.10 (K-), 2.32±0.18 (K+), 1.21±0.28 (B). One-way ANOVA test showed a significant difference between all groups after orange water kefir intervention, with the p-value <0.05. Conclusion: The antioxidative activity of orange water kefir has been proven to improve oxidative stress and fraction area of degeneration and necrosis in the kidney tissue of the hyperlipidemic rat model. |
